Myostatin (also known as growth differentiation factor 8, abbreviated GDF-8) is a myokine, a proteinproduced and released by myocytes that acts on muscle cells' autocrine function to inhibit myogenesis: muscle cell growth and differentiation.
What Is GDF-8 Peptide
Growth differentiation factor 8 (GDF8) and its antagonist follistatin-like 3 (FSTL3) are expressed in the placenta during early gestation. These two factors may play a role in the regulation of normal placentation. However, whether GDF8 regulates the expression of FSTL3 in human trophoblast cells remains to be elucidated. In this study, we aimed to investigate the effect of GDF8 on the expression of FSTL3 and the molecular mechanisms behind it, using human trophoblast cells as a research model.
Our results show that GDF8 significantly upregulates the expression and production of FSTL3, which further promotes the cytoinvasiveness of immortalized extracolonic cells and primary extracolonic cells, which are derived from the human first-trimester placenta. Furthermore, using siRNA-mediated knockdown approaches, we found that this regulatory effect is likely mediated by signaling pathways induced by ALK5-Sma- and Mad-associated protein (SMAD) 2/3. These findings deepen our understanding of the functional role of GDF8 and FSTL3 in regulating trophoblast invasiveness.
